Cellular Respiration In this lab we first used tweezers and a dropper to soak a cotton ball with KOH, then covering the cotton ball with a layer of nonabsorbent cotton. Cellular respiration uses O2 but it also produces CO2 at the same rate. As a result we used KOH to absorb the CO2 so the volume of gas in the chamber was not affected by CO2. Therefore the volume of gas decreased as the O2 was used up. We then used a graduated cylinder and filled it with 50 ml H2O, furthermore we dropped 25 germinating peas determining the water displacement, doing the same for dry peas. We also created a glass bead chamber the reason for doing this is because the glass beads will not undergo respiration. Therefore the glass beads in the respirometer are used as a control in which no changes should occur in CO2 and O2 levels. We then sealed the chamber with the pipette, placed a long piece of masking tape over the water bath and placed the chambers in the water. We then allowed the chambers 7 minutes to acclimate. After the 7 minutes we took food coloring and pushed a little amount of liquid to the tip, touching the food coloring to the pipette tip. Next we lowered the pipettes in the water bath and let them equilibrate for 3 minutes. The purpose of the equilibration in the water baths was so that the respirometers were all the same temperature. We then recorded the data, taking reading again every 3 minutes fir 12 minutes total. Lastly, we calculated the cumulative volume of O2 gas taken in over the 12 minutes and the cumulative volume difference of the glass beads. For the variable experiment we changed the organism and placed it in cold temperature rather than room temperature. (seed experiment) If groups of germinated, non-germinated, and glass beads with equal volumes are placed in respirators in water baths of 10C and 22C, measuring the consumption of oxygen, then the peas/beads in warmer temperatures will consume more oxygen (indicating a higher rate of cellular respiration) than colder temperatures. Background The equation for cellular respiration is C6H12O6 + 6O2+ 36-38 ADP+ 36-38 Pi---- 6CO2 + 6H2O + 36-38 ATP, meaning that the reactants are glucose and oxygen and yield the products carbon dioxide, water, and ATP. The key product from this reaction is ATP which serves as the source of energy used for cellular activity within the organism. The CO2 released during cellular respiration serves as a clean gas product which can be recycled into the process of photosynthesis. The product H2O is also a clean product that is used as a reactant in photosynthesis.
